Infinium MethylationEPIC&Hydroxymethylation oxEPIC アレイは、選択的な酸化、バイサルファイト処理とIllumina®技術に基づくゲノムワイドのDNAメチル化解析技術です。これは、一塩基分解能を有するゲノム全体にわたる850,000を超えるヒトCpG位置のメチル化レベル(5mC)と総メチル化(5mC + 5hmC)レベルの定量的な検出を可能にします。
バイサルファイト変換で、2種の5-メチルシトシンと5-ヒドロキシメチルシトシンのが判別なくシトシンとして読み取られ、DNAサンプルの総メチル化(5mC + 5hmC)レベルのマップが検出されます。 最初に酸化ステップを行うことで、5-ヒドロキシメチルシトシンは5-ホルミルシトシンに変換され、その後、バイサルファイト処理中にウラシルに変換されます。 その結果、5メチルシトシンのみがシトシンとして読み取られ、DNAサンプルのメチル化(5mC)レベルのマップが検出されます。 この2つの方法を組み合わせると、DNAサンプルの5-ヒドロキシメチルシトシン(5hmC)の状態を調べることが可能です。
